Study on the Mechanism of Zhenwu Decoction in Decreasing the Fats in Non-Alcoholic Fatty Liver Disease Using System Pharmacology and Cytological Validation Test

Purpose of this study was to screen the active compounds Zhenwu decoction and their gene targets in treatment non-alcoholic fatty liver disease by network pharmacology cytological validation test. The candidate related were obtained from traditional Chinese medicine system database PharmMapper. disease-related genes online Mendelian inheritance man, GeneCards DisGeNET databases. Molecular docking carried out simulate binding affinities between potential core key target genes, chemical constituents analyzed identified Ultra-Performance Liquid Chromatography-Quadrupole Time-of-Flight Mass Spectrometry technology. Therapeutic effect mechanism on validated using vitro analysis. protein-protein interaction analysis albumin, protein kinase B1, epidermal growth factor receptor, caspase 3 peroxisome proliferator activated receptor gamma as genes. Gene ontology annotation showed that decoctionnon- alcoholic mainly involved response steroid hormone lipid catabolic process. results Kyoto encyclopedia genomes pathway enrichment Forkhead box O signaling phosphatidylinositol 3-kinaseprotein B pathway. Paeoniflorgenone had highest for gamma. experiment 19.5 mg/ml (p≤0.05) 39 (p≤0.01) could reduce accumulation. Real-time quantitative polymerase chain reaction analyses revealed induced down-regulation messenger ribonucleic acid levels up-regulation heat shock 90 alpha family class a member 1, mitogen-activated 1 3-kinase levels.
